Abstract

Background and Objective: The synthesis of uncoupling protein 1 (UCP1) in adipose tissue plays an important role in providing resistance and prevention of fat accumulation, weight gain and obesity. This study was done to evaluate the effect of eight weeks aerobic training with moderate and high intensities on serum levels of Irisin and UCP1 white adipose tissue in obese male rats.
Methods: In this experimental study, 24 adult obese male Wistar rats (weight: 250 to 300 gr, BMI>30g/cm2) were randomly assigned into three groups including moderate aerobic training intensity, high intensity aerobic training and control group. The aerobic exercise training was included 8 weeks (5 sessions/per-week for 60 min per session). All training groups carried out aerobic training with 28 m/min (moderate intensity), aerobic training with 34 m/min (high intensity) on treadmill. 48 hours after the training period, the level of UCP1 and Irisin protein was measured.
Results: The level of UCP-1 in adipose tissue and serum Irisin in both aerobic training intensities increased compared to control group, but this increase only in aerobic training group with a moderate intensity was singnificant (p<0.05).
Conclusion: Eight weeks aerobic training with moderate intensity leads to increase of UCP-1 in adipose tissue and Irisin levels.

Abstract

Background and Objective: Todays, coronary artery disease is one of the most important health issues. Antioxidants are the agents that can play important role in reducing cardiovascular diseases. The present study was done to determine the effect of resveratrol supplementation during rehabilitation exercises training on systemic inflammation factors in patients after coronary artery bypass surgery.
Methods: This clinical trial study was done on 40 patients after coronary artery bypass surgery in the heart rehabilitation center of Farshchian cardiovascular specialist hospital in Hamadan, Iran during 2016. Patients randomly assigned into four groups including control (Con; n=10), resveratrol supplementation (RS; n=10), rehabilitation exercise training (RXT; n=10), and resveratrol supplements-cardiac rehabilitation exercise training (REX; n=10). Rehabilitation exercise training was done in nonconsecutive three sessions for eight weeks in the form of endurance training at 50-70% HRmax for 15-20 min, and resistance training at 20-50 one-repetition maximum (1RM). The number of repetitions was 8-15 reps, where done based on the capacity and ability of the subject, while the subjects in supplement groups consumed daily 400 mg of resveratrol. Blood sample was done before and 48 hours after exercise training and supplementation, and concentrations of Tumor necrosis factor alpha (TNFα), Interleukin- 6 (IL-6), Interleukin 1 beta (IL-1β) and C-reactive protein (CRP) were measured in the plasma.
Results: CRP, TNF-alpha and IL-1beta and IL-6 were significantly reduced in resveratrol supplements-cardiac rehabilitation exercise training group in compared to control, resveratrol supplementation and rehabilitation exercise training groups (P˂0.05).
Conclusion: This study showed that combination of consuming resveratrol with rehabilitation exercise training in reducing some inflammatory factors was evaluated selectively, more effective than rehabilitation exercise training and resveratrol supplementation alone.

Abstract

Background and Objective: Physical exercise and nutrition supplements are recommended interventions to reduce cachexia. This study was conducted to determine the effect of resistance training and resveratrol supplementation on muscle regeneration indices of MyoD (myoblast determination protein) and eMHC (embryonic Myosin Heavy Chain) in CT-26 colon cancer mice.
Methods: This experimental study was performed on 20 six-week-old BALB/c mice to which CT-26 tumor was implanted. The mice were divided into four groups of resistance training, resveratrol, combination of resveratrol with resistance training, and control. The resistance training group performed six weeks of progressive resistance training. The resveratrol group received 100 mg/kg resveratrol per day, and the control and resistance training group received the solution of Methyl cellulose through gavage. In gastrocnemius muscle MyoD protein and eMHC level were measured using western blot and ELISA methods, respectively.
Results: eMHC protein in combination of resveratrol with exercise group (4.66±0.25) increased significantly compared to the exercise group (3.46±0.64) (P<0.05). Body weight of mice without tumor (21.50±1.30) in the resistance training group increased significantly compared to other groups (P<0.05). There was no significant difference in the tumor weight of mice and MyoD protein in expermental groups.
Conclusion: Regarding the increasing effect of combination of resveratrol with exercise group compared to resistance training group on eMHC, resveratrol supplementation at higher doses may be recommended along with resistance training to improve muscle regeneration.

Abstract

Background and Objective: Many studies have been done on the effects of ginseng and green tea on inflammatory factors and liver enzymes, but no research has yet studied the comparative effects of ginseng and green tea extracts with Pomeol Breuler on C-reactive protein (CRP) and liver enzymes in hyperlipidemic rats. This study was done to evaluate the effects of ginseng and green tea extracts in comparison with Pomeol Bruler on CRP and liver enzymes in hyperlipidemic rats.
Methods: In this experimental study, 42 male Wistar rats were randomly allocated into 7 groups. Animals in group 1 (control group) received a normal diet. The experimental groups 2-7 received a high-fat diet for a month. The groups were treated with the extract for 8 weeks and by intraperitoneal injection. Groups 1 and 2 received 77.5 mg/kg/day and 155 mg/kg/day green tea extract, respectively. Groups 3 and 4 received 103.3 mg/kg/day and 206.6 mg/kg/day ginseng extract, respectively. Group 5 received 0.16 g/kg/day of Pomeol Bruler, and group 6 received green tea extract (155 mg/kg/day) and ginseng extract (206.6 mg/kg/day). At the end of the treatment, the level of CRP and liver enzymes including aspartate transaminase (AST) and alanine transaminase (ALT) were measured.
Results: The level of CRP was significantly reduced in the rats treated with ginseng, green tea, and Pomeol Bruler compared with the control group (P<0.05). The level of AST and ALT did not differ significantly between the ginseng, green tea, Pomeol Bruler, and control groups.
Conclusion: The combined use of ginseng and green tea with Pomeol Breuler for 8 weeks may reduce inflammatory factors but does not affect liver enzymes.

Abstract

Background and Objective: Ventilator-associated pneumonia (VAP) is an important complication in intensive care unit (ICU) patients with endotracheal intubation. Finding potential early indicators of this condition can aid in reducing the disease burden. We aimed to investigate the relationship between VAP occurrence and serum levels of C-reactive protein (CRP), procalcitonin (PCT), and hemoglobin (Hgb) during ICU hospitalization of brain trauma patients.
Method: This descriptive-analytical study was carried out on brain trauma patients (99 male, 39 female), referring to the 5th Azar hospital in Gorgan, Iran, in the course of 2017. The patients were hospitalized in ICU with endotracheal intubation and mechanical ventilation. The diagnosis of VAP was confirmed by a 12-score rating based on chest radiographs, body temperature, white blood cell count, and sputum culture. After admission, serum CRP, PCT, and Hgb were documented daily for 6 days.
Results: According to the diagnostic criteria, VAP was confirmed in 41 patients (30%). Serum CRP and PCT levels on the 6th-day post-admission were significantly associated with VAP diagnosis, while Hgb levels did not differ significantly between VAP and non-VAP patients. Serum levels of CRP, PCT, and Hgb on the 6th day were not associated with age or sex.
Conclusion: Our results indicate that serum CRP and PCT levels are associated with the occurrence of VAP in ICU patients receiving mechanical ventilation. Therefore, these biomarkers could be utilized to warn physicians about the possibility of VAP, thereby reducing mortality rate and hospitalization length.

Abstract

Background and Objective: Understanding the cellular signaling mechanisms involved in muscle hypertrophy is considered a scientific challenge. Mammalian target of rapamycin (mTOR) is one of the regulatory factors in this process that increases protein synthesis in skeletal muscle through phosphorylation. This study aimed to determine the effect of six weeks of high-intensity interval training (HIIT) on phosphorylated mTOR protein in the quadriceps muscles of adult male Wistar rats.
Methods: In this experimental study, 16 adult male Wistar rats (six weeks old and weighing an average of 190.93±4.97g) were used. The animals were randomly divided into two groups of control and training (n=8). The training group underwent six weeks of HIIT on a treadmill, with five sessions per week. The load was increased during the six weeks from repeating the interval of 30 meters per minute for 30 seconds in the first sessions to eleven repetitions of the interval of 35 meters per minute for 30 seconds at the end of the sixth week, with rest intervals between the intervals at a speed of 13 meters per minute for 60 seconds. The control group did not undergo any training. The mice were anesthetized, and the Vastus lateralis of the quadriceps muscle was extracted. The level of phosphorylated mTOR protein in the quadriceps muscle was measured using the immunohistochemical method.
Results: HIIT significantly increased the levels of mTOR phosphorylation protein in male Wistar quadriceps femoris muscle compared to the control group (P<0.05).
Conclusion: Interval activity can have a positive effect on muscle hypertrophy through mTOR.

Abstract

Background and Objective: Alkaline phosphatase, BMP, and GATA proteins are important factors in the process of spermatogenesis. This study aims to investigate the effect of alkaline phosphatase, GATA, and BMP expression on spermatogenic stem cells, embryonic cells, and embryonic stem-like cells (ES-like) in C57BL mice.
Methods: In this experimental study, spermatogonial stem cells were isolated from three heads of 4-week-old C57BL mice, and embryonic stem cells and ES-like cells were prepared. Alkaline phosphatase staining test was performed on spermatogenic stem cells, embryonic cells, and ES-like cells. The expression of BMP and GATA genes was analyzed using Fluidigm PCR. Protein-protein interaction networks were isolated and drawn using databases.
Results: Positive alkaline phosphatase expression in stem cells and negative expression in testicular Sertoli cells indicated the presence of this enzyme in pluripotent cells. The gene expression of BMP and GATA in spermatogonial stem cells (6.3 and 2.7, respectively), embryonic cells (3.2 and 4.4, respectively), and ES-like cells (8.5 and 2.5, respectively) was positive, but not statistically significant. Bioinformatics studies showed the regulatory role of these genes and their direct effect on alkaline phosphatase.
Conclusion: BMP and GATA genes, along with alkaline phosphatase enzymes, play a crucial role in controlling embryonic and spermatogonial stem cells, maintaining their pluripotency, and guiding them towards differentiated cells.
