fa مقایسه پارامترهای اسپرم در روش Swim up با یک و دوبار شستشو Comparison of sperm parameters in swim up method with single and double washing عمومى General تحقيقي Original Articles زمینه و هدف : روش‌های مختلفی برای بهبود پارامترهای اسپرم وجود دارد که از آن جمله می‌توان به روش Swim up اشاره کرد. شستشوی اسپرم مرحله‌ای در این روش ضروری می‌باشد. به نظر می‌رسد؛ دوبار شستشو به‌ویژه در نمونه‌هایی با غلظت بالا و مواردی که سیمن به‌خوبی مایع نمی‌شود؛ نتایج بهتری به‌دنبال داشته باشد. این مطالعه به منظور مقایسه پارامترهای اسپرم در روش Swim up با یک‌بار و دوبار شستشو انجام شد.   روش بررسی : این مطالعه آزمایشگاهی روی 50 نمونه اسپرم مرد مراجعه کننده به مرکز ناباروری حضرت فاطمه الزهرا (س) دانشگاه علوم پزشکی بابل، طی سال 1386 صورت گرفت. نمونه‌ها ‌دارای حجم 5-2 میلی‌لیتر، تعداد بیش از 20 میلیون، حرکت بالای 50درصد و مورفولوژی طبیعی بیشتر از 14درصد برابر معیار کروگر بودند. با در نظر گرفتن شرایط نمونه‌گیری، شستشوی اسپرم در طی دو مرحله صورت گرفت. داده‌ها توسط نرم افزار SPSS-10 و با استفاده از آزمون measurement repeated و Paired T-test  آنالیز شدند و P<0.05 معنی‌دار تلقی گردید.   یافته‌ها : میانگین حجم مایع سیمن در افراد مورد مطالعه 4.05±1.006 بود. میانگین حرکت اسپرم در یک‌بار و دوبار شستشو نسبت به قبل از شستشو افزایش یافت (P<0.05)، ولی بین یک و دوبار شستشو این افزایش از نظر آماری معنی‌دار نبود. با انجام شستشوی دوم، حرکت اسپرم به صورت معنی‌داری در گرید III و IV افزایش یافت و گرید I نیز کاهش پیدا کرد (P<0.05). میانگین مورفولوژی طبیعی اسپرم‌ها با شستشو افزایش یافت، لیکن بین یک بار و دوبار شستشو این افزایش معنی‌دار نبود.   نتیجه‌گیری: نتایج این مطالعه نشان داد که Swim up روش مناسبی برای آماده‌سازی اسپرم می‌باشد و دوبار شستشو سبب بهبود قابل توجه پارامترهای اسپرم می‌گردد. Background and Objective: There are different methods to improve sperm parmeters including swim up.Sperm washing is a basic step in This method during 2007. It seems that two times washing cause bether result especially in high viscosity sample and sample it is not liqufy. The aim of this study is to compare the single and double washing swim up method on sperm parameters. Materials and Methods: This study was carried out on 50 semen samples referred to Fatemeh-Alzahra infertility center of Babol University of Medical Sciences during 2007. Semen samples had the following characteristics: volume 2-5ml, count>20 million/ml, motility>50% and normal morphology>14%. Sampling condition were completely considered. Sperm parameter analysis carried out after two times washing. Data entered to SPSS-10 and analyzed by repeated measurement and paired T-Test and p-value<0.05 considered statistically significant. Results: Semen volume mean was 4.050±1.006. Mean sperm motility in one and two times washings increased (P<0.05), but there was not statistically significant differences between one and two times washing. In grading analysis, by increasing the washing, grade III, IV increase and grade I decrease (P<0.05). Sperm morphology mean increased with washing but it is not statistically different between one and two times washing.
